Animal Models and IL10

The study of animal models of colitis has demonstrated that disruption of regulatory pathways within the mucosal immune system are involved in the pathogenesis of intestinal inflammation. Of particular relevance to this chapter are models that target IL-10 signalling and the CD4+ CD45RB lymphocyte transfer model of colitis.

Leukocyte Scan

Figure 2. Leukocytic infiltration in active Crohn's disease. Intestinal inflammation in Crohn's disease is associated with an influx of leukocytes within the mucosa. A) A 99tehnetium labelled leukocyte scan from a patient with active disease. In this study, leukocytes are labelled with 99Technetium ex vivo prior to reinjection into the patient, where they are taken up by areas of inflammation (arrows) and the spleen (*). B) A histological section of the mucosa in active Crohn's disease demonstrating the classical features of partial crypt destruction, a mononuclear cell infiltrate and granuloma formation (+).
